چکیده

A spectrophotometric method was used to follow the oxidation of retinol by preparations of cattle retinas. Two-tenths millimole chlorpromazine (CPZ) and thioridazine (TRDZ) activated the initial rate. This was followed by inhibition. These effects are reversible. The ring sulfoxide of chlorpromazine and thioridazine thiomethyl sulfoxide (TPS-23) had no apparent effect. Lower concentrations (0.01 and 0.1 mM.) of CPZ and TRDZ have no demonstrable effect on the standard reaction, and no differences between CPZ and TRDZ could be shown. Chloroquine (0.2 mM.) and sodium lauryl sidfate (1.0 mM.) were shown to activate the reaction without changing the pseudomonomolecular characteristic of the standard reactions. These data were discussed with regard to the known surface activities and previously demonstrated enzyme inhibitory effects of the drugs.

Studies on the properties of retinal alcohol dehydrogenase from the rat.

An NAD-dependent alcohol dehydrogenase (alcohol : NAD oxidoreductase; EC 1.1.1.1) has been isolated and partially purified from the retinal cytosol of the rat. Its substrate specificity and sensitivity to inhibitors of hepatic alcohol dehydrogenase have been investigated. Ethanol, 1-propanol and 1-butanol served as substrates for this enzyme but the K,,, values were more than 100-fold higher th...

Retinol Dehydrogenases Regulate Vitamin A Metabolism for Visual Function

The visual system produces visual chromophore, 11-cis-retinal from dietary vitamin A, all-trans-retinol making this vitamin essential for retinal health and function. These metabolic events are mediated by a sequential biochemical process called the visual cycle. Retinol dehydrogenases (RDHs) are responsible for two reactions in the visual cycle performed in retinal pigmented epithelial (RPE) c...

Stereoisomeric specificity of the retinoid cycle in the vertebrate retina.

Understanding of the stereospecificity of enzymatic reactions that regenerate the universal chromophore required to sustain vision in vertebrates, 11-cis-retinal, is needed for an accurate molecular model of retinoid transformations. In rod outer segments (ROS), the redox reaction involves all-trans-retinal and pro-S-NADPH that results in the production of pro-R-all-trans-retinol. A recently id...

Stimulation of retinoic acid production and growth by ubiquitously expressed alcohol dehydrogenase Adh3.

Influence of vitamin A (retinol) on growth depends on its sequential oxidation to retinal and then to retinoic acid (RA), producing a ligand for RA receptors essential in development of specific tissues. Genetic studies have revealed that aldehyde dehydrogenases function as tissue-specific catalysts for oxidation of retinal to RA. However, enzymes catalyzing the first step of RA synthesis, oxid...

RDH10 is essential for synthesis of embryonic retinoic acid and is required for limb, craniofacial, and organ development.

Regulation of patterning and morphogenesis during embryonic development depends on tissue-specific signaling by retinoic acid (RA), the active form of Vitamin A (retinol). The first enzymatic step in RA synthesis, the oxidation of retinol to retinal, is thought to be carried out by the ubiquitous or overlapping activities of redundant alcohol dehydrogenases. The second oxidation step, the conve...
